Def. matters. If you go up a gauge, say from 9's to 10s or 11's, the nut may pinch the strings and cause tuning issues.
Going the other way, 10's down to 9's or 8's, the nut slots will get a bit loose, and that can cause a weird vibration on open strings as they vibrate in the nut. However, I think that a slightly wider nut slot feels quite slinky and good for some reasons.

The general theory would be chuck the new string on and see how it feels, it might be fine, it might need a tickle up

yes a regular tickle up is wise, different folks prefer a different timeframe between tickle ups but a couple of times a week is probably a good inverval
